What is a peptide?

What is peptide science?
Peptide science studies amino-acid sequences and related molecules across chemistry, structure, synthesis, receptor biology, cellular signaling, analytical characterization, and experimental pharmacology. Different questions require different methods and levels of evidence.
How does the Peptide Science Academy organize research compounds?
Profiles are grouped by broad research area and then described by molecular identity, proposed mechanism, research focus, evidence maturity, and a link to scientific literature. Related non-peptide compounds are labeled rather than silently classified as peptides.
What does an evidence-maturity label mean?
It summarizes the general stage of published research for that compound. Human clinical indicates controlled human research exists; early human indicates smaller or earlier-stage work; preclinical-heavy indicates that laboratory and animal research dominate; limited evidence identifies a narrow or weakly replicated literature base.
Are all compounds in the library peptides?
No. The Academy includes peptide analogs, hormones, bioregulators, blends, and selected related compounds that appear in the same research landscape. Profiles identify non-peptide small molecules and other classes directly to preserve chemical accuracy.
Where do the scientific references lead?
Profile links point to scientific-literature searches or primary publications so readers can inspect the underlying research. A literature link supports further investigation; it does not mean every claim associated with a compound has been clinically established.

01 · Foundations
From amino acids to a peptide chain.
Peptides are compounds formed when amino acids are connected through amide linkages. The sequence is written from its amino end toward its carboxyl end.
The peptide bond
When two amino acids form a peptide bond, the carbonyl carbon of one becomes covalently linked to the nitrogen of the next. The elements of one water molecule are lost for every bond formed in a simple linear chain.
A sequence calculation therefore adds the atoms of its amino acids, then subtracts water for each connection. Terminal groups and structural modifications must be stated separately.
02 · Process
How a synthetic sequence becomes material.
Modern solid-phase peptide synthesis assembles a protected sequence step by step on an insoluble support. The crude product still requires separation and characterization.
Synthesis
Repeated deprotection, coupling, and washing cycles extend the chain. Cleavage releases the assembled sequence from the solid support and removes specified protecting groups.
- Sequence assembly is directional and iterative.
- Incomplete coupling and side reactions can create related impurities.
- A theoretical sequence does not establish the composition of a real sample.
Purification & drying
Chromatographic purification separates components under a defined method. Lyophilization removes frozen solvent under reduced pressure to produce a dry matrix.

03 · Analytical evidence
Different questions need different methods.
Chromatography and mass spectrometry are complementary. Strong interpretation states what each method measured and avoids turning one result into several unsupported claims.
HPLC / UHPLC
Liquid chromatography separates components as they interact with a stationary phase and a moving liquid phase. A chromatogram records detector response over retention time.
- Useful for separation and relative peak-area assessment.
- Results depend on column, gradient, detector, integration, and sample conditions.
- A prominent peak is not, by itself, molecular identity confirmation.
Mass spectrometry
Mass spectrometry measures ions according to mass-to-charge ratio. Comparing observed signals with a sequence-derived mass can support identity and reveal certain variants.

Amino-acid residue
The portion of an amino acid that remains after it becomes part of a peptide chain. Peptide-bond formation removes the elements of water between adjacent amino acids.
Primary structure
The covalent amino-acid sequence of a peptide, conventionally written from the N-terminus to the C-terminus. Modifications and disulfide connectivity are separate structural details.
Theoretical molecular mass
A calculated mass derived from a stated sequence and explicit modifications. An observed mass may differ because of salts, adducts, isotopes, counterions, hydration, or unmodeled modifications.
Isoelectric point (pI)
The pH at which a molecule’s modeled net charge is approximately zero. A calculated pI depends on the pKa set and assumptions used.
Chromatographic purity
The relative area assigned to a target peak under a particular chromatographic method and detection setting. It is not automatically the same as peptide content, identity, or total material purity.
Mass-to-charge ratio (m/z)
The quantity measured for an ion in mass spectrometry. Charge state, ionization, adducts, and instrument conditions matter when comparing a spectrum with a theoretical neutral mass.
